Over the years i have done many combo's of auto trannys. Auto's with trans brakes & air operated rpm shifters they can't be beat for consistant bracket racing. But the money invested is high.
TH400's can be made to handle 1500+ hp in heavy cars.
I have a high end dollar 700R4. My next major buy will be a manual 5 or six speed just to save the 25-40 hp that an auto tranny uses.
Trans braked car's are so easy to get good lights with high rpm launches. If your into drag racing only a reverse paterned full manual shifted is hard to beat.
I have had as much as a 6500 stall air shifted rpm activated shifter. Then it's just the throttle stop and engine tune to right on a number for class racing

I have a richmond 5 speed behind my 502 with a 3.08 rearend. I kind of like that combo. When I got the car, it had a 3.70 rearend and the 1st gear was a super granny gear and I would launch my car in 2nd gear at the drag strip.

Go with the ROD, I have one and it handles more power than you are putting out I have flogged mine with 625 rwhp on a chassis dyno. If you are a dedicated drag racer go with the auto, if not go with the 6 spd.

If you want to go fast, get the 6 speed, if you want to be consistant, get an automatic. A manual will always be quicker.

The richmond 6 and 5 speed trannys are rated for 450 tq and the ZZ502 has around 550 tq. A lot of guys run them without problems and if you don't run slicks it shouldn't be a problem.
BowTie makes a 700R4 rated up to 630 tq.
Your title for this post "Best tranny behind a ZZ502", IMHO would be a heavy duty th400 with a Gear Vendors (GV) overdrive unit behind it. With the GV unit it would actually give you a 6 speed auto.

Im running a 35 year old M-21 4 speed, and 35 year old 3:55 rear end on my 502. Keep in mind, this motor is rated (the 502/502) at I beleive 560 ft pds of tourque. With my Edlebrock EFI, Id have to go a little higher
Ive wrung my 502 out more than once, and have yet to encounter a prob, BUT I was very fortunate, as myself, and folks in the past that might have owned my 68 (original 427 car) seemed to have really took the time to ensure proper maintainance and oil changes, two types of oil changes we tend to forget (tranny/rear end).
While there are for sure, better trannies out there, my stock M-21 and rear end are doing fine so far
