No you are fine replacing just one. No matter how many you replace though you will need to resync them to the car which is only a 2 minute process for someone with a tool.

Hey Bob. Can you get the early 05's that can be activated to be programmed by changing air pressure but still work on the 05-09 systems?

I think you're talking about the sensors used in the 2004 Cadillac XLR and the very early 2005 XLR and C6 (built before about Aug 2004).
They will work in all 2005-2009 C6 Vettes made for the U.S./Canada.
First of all, they could be be triggered by a magnet, but not by changing air pressure.
Second of all, they haven't been made for about 7 or 8 years, so nobody has any - unless you find some old ones on eBay.
