The hole referred to by the OP is an oil passage. Not all small blocks have this passage drilled. Many are drilled & tapped for a 1/4" pipe plug. on some, it was factory plugged with a 1/2"OD "cup" type plug... essentially a little freeze plug, but not a "freeze plug".
Here is a pic from a 1965 327 L79 that I am currently building.
I also have a 300HP 1966 327 where that hole was never drilled. And I have two 350 4bolt main blocks where it is drilled/tapped and plugged with a 1/4" pipe plug.

If yours has threads in it then it may have been tapped by someone for a pipe plug- but it should be 1/4" NPT if so.

ANY engine that I'm installing in a car first goes on my home-made engine stand for a run. Water and oil leaks, timing and valve adjustment are easily addressed before the installation... Al
